RESEARCH
Alzheimer’s Disease (AD) is a progressive neurodegenerative disorder affecting over 12 million people worldwide. Post mortem autopsy of an AD patient’s brain reveals two characteristic features: senile plaques and neurofibrillary tangles. The senile plaques, which are unique to AD, consist of aggregated beta-amyloid peptide, which is derived from the amyloid precursor protein (APP). A small number of patients with familial Alzheimer’s disease have mutations in APP. Because of the difficulty of investigating the role of APP in mammals, we are using a simple model system, the worm C. elegans, to determine the role of APP-related proteins. The function of related proteins are generally conserved through evolution. In C. elegans, a single gene, apl-1, encodes a protein similar to human APP. Loss of apl-1 results in larval lethality, indicating that apl-1 plays an essential role in postembryonic development. This lethality can be rescued by microinjection of a wild-type copy of apl-1. By using transgenic strains that overexpress APL-1 in different cells of the animal, we can modify the lifespan of these animals. We are investigating the basis of the lifespan effects on apl-1 expression.
